Effect of paclitaxel (Taxol®) on the biodistribution of sodium pertechnetate (Na99mTcO4) in female Wistar rats BABT
Holanda,Cecília Maria de Carvalho Xavier; Oliveira,Elias Herculano de; Rocha,Louisianny Guerra da; Barbosa,Vanessa Santos de Arruda; Spyrides,Maria Helena Constantino; Aragão,Cícero Flávio Soares; Medeiros,Aldo da Cunha.
The evidence that natural or synthetic drugs can affect the biodistribution of radiopharmaceuticals (radiobiocomplexes) in setting of nuclear medicine clinic is already known. We studied the effect of Paclitaxel, an anti-neoplastic agent for the treatment of solid tumors, on the biodistribution of Na99mTcO4 in female rats. Paclitaxel (1mg/mL/week) was administered into animals in single dose during 3 weeks, with interval of 1 week among them. The control group received NaCl 0.9% solutions by the same via. One hour after the last dose, it was injected Na99mTcO4 in the animals. The percentage of activity per gram (%ATI/g) and biochemical and hematological determinations were performed. A significant increase were found in alanine aminotransferase, aspartate...

Lipid core nanoparticles resembling low-density lipoprotein and regression of atherosclerotic lesions: effects of particle size BJMBR
Freitas,S.C.M.P.; Tavares,E.R.; Silva,B.M.O.; Meneghini,B.C.; Kalil-Filho,R.; Maranhão,R.C..
Particles are usually polydispersed and size is an important feature for lipid-based drug delivery systems in order to optimize cell-particle interactions as to pharmacologic action and toxicity. Lipid nanoparticles (LDE) with composition similar to that of low-density lipoprotein carrying paclitaxel were shown to markedly reduce atherosclerosis lesions induced in rabbits by cholesterol feeding. The aim of this study was to test whether two LDE fractions, one with small (20–60 nm) and the other with large (60–100 nm) particles, had different actions on the atherosclerotic lesions. The two LDE-paclitaxel fractions, prepared by microfluidization, were separated by density gradient ultracentrifugation and injected (4 mg/body weight, intravenously once a week)...

Paclitaxel and spirulina co-loaded polymeric nanoparticles: in-vitro and in-vivo anticancer study BJPS
Ravikumar,Yamuna; Catherine,Dorcas Pinky; Nair,Keerthi G. S; Sekar,Akshya; Velmurugan,Ramaiyan.
Paclitaxel spirulina nanoparticles were said to have promising anticancer activity against gastric cancer. Nanoparticles of paclitaxel-spirulina were prepared for treating gastric cancer using precipitation technique. The synergistic anticancer efficiency againstMKN45 cells retains when the paclitaxel and spirulina were encapsulated into nanoparticles. To increase the site specific delivery, intra-tumoral administration was carried in the in vivo evaluation. There was an increase in overall survival in an MKN45-transplanted mice model and notable improvement in anti-tumour efficacy when paclitaxel-spirulina nanoparticles were delivered through intra-tumoral administration. The further investigation of overall anticancer mechanism of these nanoparticles is...

Screening of endophytic fungi stored in a culture collection for taxol production BJM
Andrade,Hortência Farias de; Araújo,Lívia Caroline Alexandre de; Santos,Bruno Souza dos; Paiva,Patrícia Maria Guedes; Napoleão,Thiago Henrique; Correia,Maria Tereza dos Santos; Oliveira,Maria Betânia Melo de; Lima,Gláucia Manoella de Souza; Ximenes,Rafael Matos; Silva,Túlio Diego da; Silva,Girliane Regina da; Silva,Márcia Vanusa da.
Abstract In this work, four isolates of endophytic fungi (Alternaria alternata, Colletotrichum gloesporioides, Glomerella cingulata and Nigrospora sphaerica), deposited in the culture collection ‘University Recife Mycologia’ (URM) at the Universidade Federal de Pernambuco, were characterized for the genes ITS 1 and 4 (region 5.8 S) and evaluated for taxol production.

Synergistic effect of microRNA and albumin-bound nanoparticles for inhibition of glioblastoma cancer cell proliferation BJPS
Shariatnasery,Maria; Irani,Shiva; Soleimani,Masoud; Goodarzi,Navid; Dinarvand,Rassoul.
The functional significance of upregulation miR-34a in combination with albumin-bound paclitaxel nanoparticles in U251 glioblastoma cell line has been evaluated. The MTT assay determined that miR-34a and albumin-bound paclitaxel nanoparticles can reduce cell viability, but the combination of both factors has a stronger effect on cell viability. The application of qRT-PCR has demonstrated that the transduction of miR-34a could lead to exogenous upregulation of miR-34a level and downregulation of SURVIVIN. Moreover, treatment of U251 cells with miR-34a and nanoparticles together considerably inhibit SURVIVIN expression compared to miR-34a and nanoparticles alone. Flow cytometry showed that upon miR-34a overexpression cell cycle arrested in G1 phase, while...
